If I were to plan on tapping into my schrader valve for my wet nitrous kit on the LT1 do I need to just remove the plastic cap on the valve and screw on the fuel hose? Or do I need to pull the fuel rails off entirely and remove the valve itself? Or should I look for a different method such as tapping into the main hose with a t-fitting?
Any help would be appreciated.

You would need to remove the plastic cap, then remove the " core " from the valve, Then screw on the fitting to run the line to the nitrous solenoid.

Make sure you depressurize the system first. You dont want gas spraying all over you and your car when you pull the valve out. just pull the fuel pump fuse and run the engine till it dies.
